SLU-PP-332: A Comprehensive Overview for Biology and Medical Researchers

What is SLU‑PP‑332?

SLU‑PP‑332 (CAS 303760‑60‑3) is a small-molecule pan‑estrogen‑related receptor (ERR) agonist, particularly selective for ERRα with an EC₅₀ of 98 nM, and also active on ERRβ (230 nM) and ERRγ (430 nM) (Tocris Bioscience). As a hydrazone derivative of 4-hydroxybenzamide and 2-naphthaldehyde, SLU‑PP‑332 modulates nuclear receptor activity with high specificity.

Chemical Structure & Properties

  • Chemical Name: (E)-4-Hydroxy-N′-(naphthalen-2-ylmethylene)benzohydrazide
  • Molecular Formula: C₁₈H₁₄N₂O₂
  • Structural Features: A hydrazone linkage between a phenolic group and naphthalene moiety, with the active Z-configuration at the imine bond (Wikipedia).
  • Solubility: Soluble in DMSO (~29 mg/mL); moderate aqueous solubility may require solvent optimization (Tocris Bioscience).

SLU‑PP‑332 is a non-peptide small molecule—not applicable for amino acid sequence or peptide modifications.

 Mechanism of Action

SLU‑PP‑332 activates ERRs—especially ERRα—by stabilizing their active conformation, enhancing affinity for PGC‑1α co-activators (Wikipedia). This leads to simulations of energy-demanding states, increasing:

  • Mitochondrial biogenesis via upregulation of TFAM and OXPHOS genes
  • Fatty acid oxidation, through CPT1 and MCAD enhancement
  • AMPK pathway activation, promoting metabolic adaptation

These effects mimic physiological responses to exercise and caloric stress (Wikipedia, UF News).

 Research-Proven Benefits

In vitro studies demonstrate:

  • Enhanced mitochondrial respiration in muscle cells (e.g., C2C12 myocytes) (Amazon).
  • Upregulation of ERR target genes such as Pdk4 (Amazon).

In vivo (preclinical) findings show:

  • Reduced fat mass, improved glucose tolerance, and insulin sensitivity in metabolic syndrome models (Tocris Bioscience).
  • Increased endurance, oxidative muscle fiber composition, and running capacity (~30–40%) .
  • Mitochondrial restoration and reduced inflammation in aged kidneys and hearts via ERR agonism (AdipoGen).

Research Applications for In Vitro Studies

SLU‑PP‑332 is ideal for exploring:

  • Mitochondrial function assays, e.g., OCR/ECAR measurements
  • Metabolic gene expression, especially PGC-1α, TFAM, CPT1
  • Signal pathway analysis, including AMPK, via western blotting or reporter assays
  • Outcomes in muscle/myotube and hepatocyte models related to fatty acid and glucose metabolism
  • Mechanistic screening of mitochondrial activators and metabolic modulators

Future Research Directions

  • Organoid or co-culture systems to study ERR-driven metabolic crosstalk
  • Comparative pharmacology vs selective GSK4716 or XCT790 to probe receptor subtype activity
  • Proteomics and transcriptomics following compound exposure in human cells
  • Dermal cell models exploring mitochondrial or metabolic skin rejuvenation effects
  • Neuroprotective or cardiac models to evaluate ERR-mediated mitochondrial benefits
